Add on storage, the amount of IO transactions, plus bandwidth, and you end up with something more expensive than a dedicated server that works about the same but does not perform as well. If you're doing a database in EC2 and you're storing the database files in S3, that's going to be quite slow as the files would be on something that is essentially an NFS server. The only benefit is quality bandwidth and being able to bring up a new instance on demand.
